In humans, NAD+ can be synthesized from nicotinic acid, nicotinamide, nicotinamide riboside, and tryptophan through overlapping pathways. The salvage pathway recycles nicotinamide back to NAD+ and is often considered a major route in many tissues. Dietary precursors and intracellular recycling both contribute to the pool, but the quantitative importance of each source remains an active research question. NAD+ levels are not uniform across organs or cell compartments. Measurements in blood do not necessarily reflect concentrations inside tissues.
NAD+ is a dinucleotide composed of nicotinamide, ribose, and adenine linked by phosphate groups. Its full name is nicotinamide adenine dinucleotide, with "+" denoting the oxidized form. The molecule acts as a coenzyme in redox reactions, cycling between NAD+ and NADH. In cells, it participates in electron transfer during glycolysis, the citric acid cycle, and oxidative phosphorylation. It is distinct from NADP+, which carries an additional phosphate group and supports different biosynthetic reactions.
Beyond redox chemistry, NAD+ serves as a substrate for enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins, PARPs, and CD38-family enzymes consume NAD+ and produce nicotinamide and ADP-ribose-related products. These reactions link NAD+ availability to DNA repair, chromatin modification, and cellular signaling. Because the molecule is central to energy metabolism and regulation, changes in its concentration are studied in aging, immunity, and metabolic research. The balance between synthesis and consumption varies by tissue, developmental stage, and physiological state.
Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave it and attach its ADP-ribose portion to other molecules. This group includes poly(ADP-ribose) polymerases, CD38, and sirtuins. Such reactions consume NAD+ and can influence its availability for metabolism. Cells replenish NAD+ through a salvage pathway that recycles nicotinamide and through routes starting from tryptophan or vitamin B3 forms. How these synthesis and consumption routes are coordinated across tissues remains an active area of study, and compartment-specific concentrations are difficult to measure directly.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide built from adenine, nicotinamide, two ribose sugars, and two phosphate groups. The oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, is neutral. This pair acts as a reversible electron carrier in cells. NAD+ is present in bacteria, plants, animals, and fungi. Its structure allows it to accept and donate electrons without being consumed in the reactions it supports.

The nicotinamide ring undergoes reversible reduction at the para position, converting NAD+ to NADH. This reaction transfers a hydride equivalent, not a free hydrogen atom or electron alone. Because the redox pair has a defined reduction potential, it links oxidation of fuels to respiratory chain activity. Many dehydrogenases use NAD+ as a co-substrate and produce NADH. The ratio of NAD+ to NADH reflects metabolic state and influences flux through several pathways.
NAD+ also serves as a substrate for enzymes that cleave it, including sirtuins, PARPs, and CD38. These enzymes consume NAD+ and release nicotinamide and ADP-ribose or related products. The dual roles as redox cofactor and signaling substrate connect NAD+ to DNA repair, circadian regulation, and calcium signaling. Cellular NAD+ concentrations vary by tissue, time of day, and stress exposure. How these consumption pathways interact with redox balance remains an active area of research.
NAD+ is a dinucleotide composed of two nucleotides joined by a pyrophosphate linkage. One nucleotide contains adenine, and the other contains nicotinamide. The oxidized form carries a positive charge on the nicotinamide ring and is abbreviated NAD+. It functions as a cofactor in hydride-transfer reactions, accepting electrons in catabolic pathways. In cells, it interconverts with reduced NADH, forming a redox couple central to energy metabolism. The molecule is water-soluble and does not cross cell membranes freely without specific transport or precursor pathways.
Measuring NAD+ in biological samples requires rapid processing because the compound can degrade or interconvert after collection. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and mass spectrometry. Each method has different sensitivity, specificity, and susceptibility to interference from related nucleotides. Sample type matters: cultured cells, animal tissues, and human blood present distinct challenges. Reported values can vary widely across laboratories because of differences in extraction, normalization, and analytical platform. Standardization remains an open issue in the field.
NAD+ is relatively unstable in aqueous solution, especially at neutral or alkaline pH and at elevated temperatures. It is typically stored dry, protected from light and moisture, and kept cold or frozen for long-term use. Solutions are often prepared fresh or buffered to mildly acidic pH to slow hydrolysis. Repeated freeze-thaw cycles can reduce integrity. Laboratories may verify concentration using ultraviolet absorbance at 259 nm or by enzymatic assay. These handling practices are general laboratory conventions rather than universal rules.
However, Watson and Crick found fault in her steadfast assertion that, according to her data, a helical structure was not the only possible shape for DNA—so they had a dilemma. In an effort to clarify this issue, Max Ferdinand Perutz later published what had been in the progress report, and suggested that nothing was in the report that Franklin herself had not said in her talk (attended by Watson) in late 1951. Perutz explained that the report was to a Medical Research Council (MRC) committee that had been created to "establish contact between the different groups of people working for the Council". Randall's and Perutz's laboratories were both funded by the MRC. It is also not clear how important Franklin's unpublished results from the progress report actually were for the model-building done by Watson and Crick. After the first crude X-ray diffraction images of DNA were collected in the 1930s, William Astbury had talked about stacks of nucleotides spaced at 3.4 angström (0.34 nanometre) intervals in DNA. A citation to Astbury's earlier X-ray diffraction work was one of only eight references in Franklin's first paper on DNA. Analysis of Astbury's published DNA results and the better X-ray diffraction images collected by Wilkins and Franklin revealed the helical nature of DNA. It was possible to predict the number of bases stacked within a single turn of the DNA helix (10 per turn; a full turn of the helix is 27 angströms [2.7 nm] in the compact A form, 34 angströms [3.4 nm] in the wetter B form).
=== Valve Corporation v. Vivendi Universal Games === Between 2002 and 2005, Valve was involved in a complex legal dispute with its publisher, Vivendi Universal Games (under Vivendi's brand Sierra Entertainment). Valve had entered into a publishing agreement with Sierra to release Half-Life and subsequent games in 1997, with the contract giving Sierra some intellectual property (IP) rights to Valve's games. After Valve began development of Half-Life 2, it agreed a new contract with Sierra in 2001, removing these rights from Sierra and giving Valve some rights for digital distribution. Internally, Valve started work on Steam as a means to digitally distribute these games, and first revealed this project at the March 2002 Game Developers Conference. By August 2002, Valve had found that Sierra was distributing copies of their games to Internet cafes against the terms of their contracts and filed a lawsuit against Sierra and Vivendi. In addition to claims of copyright infringement, Valve asserted that Sierra breached contract by withholding royalties and delaying the release of Counter-Strike: Condition Zero until after the holiday season. Vivendi and Sierra countersued, stating that Valve had misrepresented their position in the revised 2001 contract since they had been working on Steam at that point as a means to circumvent the publishing agreement. Vivendi sought intellectual property rights to Half-Life and a ruling preventing Valve from using Steam to distribute Half-Life 2. The countersuits, if successful, likely would have bankrupted Valve, according to COO Scott Lynch.
Peptide PHI, also known as peptide histidine isoleucine, is a peptide which functions as a hormone. This peptide contains a composition of 27 amino acids with histidine on the N-terminus and isoleucine on the C-terminus. It was originally isolated from the mammalian small intestine amongst mammalian neurons called intramural neurons which function in the motor activity of the intestinal walls. An example of this was revealed in a study that demonstrated that this peptide regulates water and electrolyte transportation in the human jejunum; similar to its inhibitory effects on fluid absorption in the small intestine of pigs and rats. Peptide histidine isoleucine (PHI) is part of family that plays a vital role in the cell growth rate such as in the intestine as well as in brain. It was derived from glucagon family called the pituitary adenylate cyclase-activating polypeptide (PACAP) and it has an amino acid sequence homology to vasoactive intestinal peptide, secretin, glucagon, and other growth hormone releasing factor. Human studies have shown that the release of PHI into the stomach regulates the neuroendocrine cell processes that affect gastrointestinal physiology. This peptide is present within the central nervous system that help regulate food consumption behavior, while at peripheral nervous system this peptide accumulates in the stomach which controls the digestion of food. It also plays a role in the regulation of prolactin in humans.
=== International === On review aggregator website Rotten Tomatoes, the film has a 93% approval rating based on review from 73 critics, with an average score of 7.70/10; the site's "critics consensus" reads: "It'll be too disturbing— and meandering— for some, but Dogtooth is as disturbing and startlingly original as modern filmmaking gets". On Metacritic, the film has a weighted average score of 72 out of 100 based on 18 critics, indicating "generally favorable reviews". Alistair Harkness of The Scotsman hailed Lanthimos as "a bold new voice on the world cinema scene, someone who might soon be elevated to a similar position as those twin pillars of Euro provocation: Lars von Trier and Michael Haneke", but added that the film is "not [...] designed simply to shock in the way von Trier's work often does, [...] nor does it have that annoyingly prescriptive, punitive air of superiority favoured by Haneke's films." Peter Bradshaw of The Guardian praised the filmmaking, finding Dogtooth "superbly shot, with some deadpan, elegant compositions, and intentionally skewiff framings". Roger Ebert of the Chicago Sun-Times gave the film three out of four stars, noting the director's "complete command of visuals and performances. His cinematography is like a series of family photographs of a family with something wrong with it.
==== Conducting Wire ==== Due to Graphene's high electrical and thermal conductivity, mechanical strength, and corrosion resistance, one potential application is in high-power energy transmission. Copper wire has long been used for power transmission for its high conductivity, ductility, and low costs. However, traditional wire fails to meet the transmission requirements of many new technologies. Thermally dependent resistivity in mesoscopic copper wire limits efficiency and current carrying capacity in small-scale electronics. Additionally, copper wire exhibits internal failure by electromigration at high current density, limiting miniaturization of wire. Copper's high weight and low temperature oxidation also limit its applications in high-power transmission. Increasing demand for high ampacity transmission in electronics and electric vehicle applications necessitate improvements in conductor technology. Graphene-copper composite conductors are a promising alternative to standard conductors in high-power applications. In 2013, researchers demonstrated a one-hundred-fold increase in current carrying capacity with carbon nanotube-copper composite wires when compared to traditional copper wire. These composite wires exhibited a temperature coefficient of resistivity an order of magnitude smaller than copper wires, an important feature for high load applications.

==== More essays on mass culture and literature ==== Adorno returned to Santa Monica to survey his prospects at the Hacker Foundation, as he would have forfeited his American citizenship by the middle of 1952 if he had continued to stay outside the country. While there, he wrote a content analysis of newspaper horoscopes (now collected in The Stars Down to Earth) and the essays "Television as Ideology" and "Prologue to Television"; even so, he was pleased when, at the end of ten months, he was enjoined to return as co-director of the Institute. Back in Frankfurt, he renewed his academic duties and, from 1952 to 1954, completed three essays: "Notes on Kafka," "Valéry Proust Museum," and an essay on Schoenberg following the composer's death, all of which were included in the 1955 essay collection Prisms. In response to the publication of Thomas Mann's The Black Swan, Adorno penned a long letter to the author, who subsequently approved its publication in the literary journal Akzente. A second collection of essays, Notes to Literature, appeared in 1958. After meeting Samuel Beckett while delivering a series of lectures in Paris the same year, Adorno set to work on "Trying to Understand Endgame," which, along with studies of Proust, Valéry, and Balzac, formed the central texts of the 1961 publication of the second volume of his Notes to Literature. Adorno's entrance into literary discussions continued in his June 1963 lecture at the annual conference of the Hölderlin Society.
== Animal models == Mice genetically engineered to lack glutathione peroxidase 1 (Gpx1−/− mice) are grossly phenotypically normal and have normal lifespans, indicating this enzyme is not critical for life. However, Gpx1−/− mice develop cataracts at an early age and exhibit defects in muscle satellite cell proliferation. Gpx1 −/− mice showed up to 16 dB higher auditory brainstem response (ABR) thresholds than control mice. After 110 dB noise exposure for one hour, Gpx1 −/− mice had up to 15 dB greater noise-induced hearing loss compared with control mice." Mice with knockouts for GPX3 (GPX3−/−) or GPX2 (GPX2−/−) also develop normally However, glutathione peroxidase 4 knockout mice die during early embryonic development. Some evidence, though, indicates reduced levels of glutathione peroxidase 4 can increase life expectancy in mice.

The glans is completely or partially covered by a double-layered fold of skin, known as the foreskin. In adults, glans exposure can be easily achieved by manual retraction of the foreskin and sometimes automatically during erection. The degree of automatic foreskin retraction varies considerably depending on the foreskin length. The foreskin can be characterized as long when the preputial orifice extends beyond the glans during erection, or medium when the orifice is located around the meatus. The primary purpose of the foreskin is considered to be the covering of the glans and the urinary meatus, while also maintaining the mucosa in a moist environment. Foreskin retractability gradually increases with age. In infancy, the foreskin is fused to the glans. It remains non-retractable in early childhood. It tightens during preadolescence. The skin begins to loosen up significantly during puberty, allowing the glans to be completely exposed when needed. By the age of eighteen, most boys will have a fully retractable foreskin. The glans on a circumcised penis remains fully exposed and dry. Generally, studies suggest that the glans remains equally sensitive in circumcised and uncircumcised penises.

== History == Providence Therapeutics Holdings Inc. was co-founded in Toronto, Ontario by Calgary, Alberta-based businessman Brad T. Sorenson and San Francisco–based Eric Marcusson Ph.D., who was also the Chief Scientific Officer. PTX-COVID19-B is a messenger RNA (mRNA)-based COVID-19 vaccine. In an interview with CTV news, Sorenson said they were "building some of the important building blocks for the messenger RNA ... that provides instructions to cells ... to build proteins that may treat or prevent disease". As of January 2021, Northern RNA's Calgary lab was proposed as the site where manufacturing of PTX-COVID19-B would take place. Providence Therapeutics' partner, Northern RNA, which located at 421 7 Avenue SW in Calgary, has been described as Providence Therapeutics northern division. A February 2021 Manitoba government press release said that the Winnipeg-based Emergent BioSolutions would be manufacturing the vaccine.

Jasmonates (JAs) are lipid-based hormones that were originally isolated from jasmine oil. JAs are especially important in the plant response to attack from herbivores and necrotrophic pathogens. The most active JA in plants is jasmonic acid. Jasmonic acid can be further metabolized into methyl jasmonate (MeJA), which is a volatile organic compound. This unusual property means that MeJA can act as an airborne signal to communicate herbivore attack to other distant leaves within one plant and even as a signal to neighboring plants. In addition to their role in defense, JAs are also believed to play roles in seed germination, the storage of protein in seeds, and root growth. JAs have been shown to interact in the signalling pathway of other hormones in a mechanism described as "crosstalk." The hormone classes can have both negative and positive effects on each other's signal processes. Jasmonic acid methyl ester (JAME) has been shown to regulate genetic expression in plants. They act in signalling pathways in response to herbivory, and upregulate expression of defense genes. Jasmonyl-isoleucine (JA-Ile) accumulates in response to herbivory, which causes an upregulation in defense gene expression by freeing up transcription factors. Jasmonate mutants are more readily consumed by herbivores than wild type plants, indicating that JAs play an important role in the execution of plant defense.

The plus sign indicates the oxidized form of nicotinamide adenine dinucleotide, which can accept electrons. When it accepts electrons, it becomes NADH. The two forms together support redox reactions in cells.
No. NAD+ is the oxidized form and NADH is the reduced form. They differ by two electrons and a proton equivalent, and cells interconvert them during metabolism.
Yes. NAD+ is present in all living cells and is required for fundamental metabolic reactions. Its concentration varies by tissue, compartment, and time.
It indicates the oxidized form, which has a positive charge on the nicotinamide nitrogen. The reduced partner NADH lacks that charge and carries added electrons. The plus sign is part of the standard abbreviation, not a separate ion.
